Anti-cancer Effects of Different Molecular Weight Chitosan Oligosaccharides in Human Colon Cancer Cells

Abstract
Here we report the anti-proliferative effect of three kinds of chitosan oligosaccharides (COSs) with relatively high molecular weight COS (HMWCOS, 5-10 kDa), medium molecular weight COS (MMWCOS, 3-5 kDa) and low molecular weight COS (LMWCOS, 1-3 kDa) in HT-29 human colon cancer cells. Among all COSs, LMWCOS exhibited the highest anti-roliferative effects in HT-29 cells. Furthermore, LMWCOS induced apoptosis in a dose-dependent manner on HT-29 cells. Treatment of cells with LMWCOS also induced the increase in caspase activity, PARP cleavage, and pro-apoptotic proteins and the decrease in anti-apoptotic proteins. In addition, inflammatory proteins were down-regulated by LMWCOS. These results indicated that the potential inhibitory effect of LMWCOS against growth of HT-29 cells might be associated with induction of apoptosis through p53 and NF-¥êB dependent pathway.
